ABSTRACT

A study to phenotypically characterize and determine the antibiogram of coagulase positive Staphylococci (CoPS) from the external surfaces of hospital cockroaches (Periplaneta americana) was conducted using standard microbiological methods. Out of the 50 cockroaches collected from various hospitals in Uyo, sixty-two percent (n = 31) had coagulase positive Staphylococci which consisted of Staphylococcus aureus (44.0 %; n = 22) and Staphylococcus intermedius (18.0 %; n = 9). The CoPS isolates showed 100% resistance to Penicillin, Tetracycline, Clindamycin and 80.6% sensitivity to Amoxicillin-clavulanate. The CoPS showed multiple antibiotic resistances to ≥ 3 antibiotics, with 60 % exhibiting resistance to 6 antibiotics. Out of the 80 % (n = 31) of the multidrug resistant CoPS that were sensitive to Amoxicillin-clavulanate, none of them showed production of beta lactamase. The cockroaches bore multiple antibiotic resistant CoPS on their external surfaces and their contact can initiate contamination of patients' food. Pest control measures in hospital are hereby recommended to minimize cockroach related infections

ABSTRACT

Salami is a ready-to-eat (RTE) product frequently purchased at street fairs in Porto Alegre. Salmonella enterica, Listeria monocytogenes, and coagulase-positive Staphylococcus (CPS) are important causes of foodborne disease and can be transmitted through the consumption of RTE foods. The aim of this study was to evaluate the presence of these pathogens in salami sold at street fairs. Ninety salami samples from three commercial brands available at street fairs were analyzed by routine bacteriological methods for Salmonella spp. and Listeria spp., as well as enumeration of CPS. In addition, two samples from each commercial brand were analyzed for water activity (aw). Samples of brand A showed aw values (0.938 and 0.942) above those set by the legislation, while brand B (0.849 and 0.860) and brand C (0.826 and 0.854) were compliant. Microbiological analyses showed that 67.7% were negative to all investigated bacteria. Salmonella Typhimurium was isolated from 4.4% (4/90) of salami samples, all from commercial brand A. ­Listeria monocytogenes was detected in 3.3% (3/90) of samples, from commercial brands B and C. Moreover, 7.7% (7/90) of samples contained CPS populations non-compliant with legislation. Although the great majority of salami sold at street fairs of Porto Alegre was compliant with standards, S. enterica, L. monocytogenes, and CPS ≥ 5 × 103 cfu.g-1 could be found in this RTE product. Therefore, control measures in the processing industry and consumer's education about foodborne illness prevention should be maintained.(AU)


Salame é um alimento pronto para o consumo frequentemente adquirido pela população em feiras livres de Porto Alegre. Salmonella enterica, Listeria monocytogenes e Staphylococcus coagulase positiva são importantes causas de doenças transmitidas por alimentos e podem ser veiculadas por alimentos prontos para o consumo. O objetivo desse estudo foi avaliar a presença desses patógenos em salames vendidos em feiras livres. Noventa amostras de salame pertencentes a três marcas comerciais foram analisados por métodos bacteriológicos de rotina quanto à presença de Salmonella spp. e Listeria spp., bem como enumeração de Staphylococcus coagulase positiva (SCP). Além disso, foi determinada a Atividade de Água (aw) de duas amostras de cada marca comercial. Amostras da marca A apresentaram valores de aw (0,938 e 0,942) acima do permitido na legislação, enquanto as amostras da marca B (0,849 e 0,860) e C (0,826 e 0,854) não violaram esse parâmetro. A análise microbiológica demonstrou que 67,7% das amostras foram negativas para todas as bactérias investigadas. Salmonella Typhimurium foi isolada de 4,4% (4/90) das amostras de salame, todas da marca comercial A. Listeria monocytogenes foi detectada em 3,3% (3/90) das amostras das marcas B e C. Além disso, 7,7% (7/90) das amostras apresentaram SCP acima do número permitido pela legislação. Apesar da grande maioria dos salames comercializados em feiras livres estarem de acordo com a legislação, S. enterica, L. monocytogenes e SCP ≥ 5 × 103 cfu.g-1 podem estar presentes nesse alimento pronto para o consumo. Dessa forma, o controle nas indústrias e a educação dos consumidores sobre a prevenção de doenças transmitidas por alimentos devem ser mantidos.(AU)

ABSTRACT: Moderate and high humidity cheeses are described as important vehicles of pathogens in many foodborne diseases outbreaks. Microbial contamination can occur in raw material or in the different steps of the product processing due to inadequate hygiene practices. Thus, the aim of this study was to evaluate the microbiological quality and safety in the production of moderate and high humidity cheese. Samples from raw milk, handlers' hands surface, final product were collected in three cheese manufacturing plants located in southern Brazil, with different levels of sanitary control. Effectiveness of milk pasteurization was also evaluated. Thermotolerant coliforms, coagulase-positive staphylococci (CPS), Salmonella spp., and Listeria monocytogenes were evaluated. Raw milk samples showed the highest contamination levels, with enumeration of 1.1x105 most probable number (MPN) mL-1 for thermotolerant coliforms, 4x105 colony-forming units (CFU) mL-1 for CPS and presence of Salmonella spp. CPS were also reported in one sample of handler's hands surface. However, only one sample of the final product was out of Brazilian regulatory standards, exceeding the limit allowed for CPS. Milk pasteurization process used in cheese preparation was effective, regardless the level of sanitary control of the industries. Results highlighted the need for better hygiene practices, in obtaining the raw milk and in the handling during the cheese manufacturing steps.


RESUMO: Os queijos com média e alta umidade são alimentos prontos para o consumo, que têm sido descritos como veiculadores de patógenos em diversos surtos de doenças transmitidas por alimentos. A contaminação microbiana pode ter origem na matéria prima, ou ocorrer durante as etapas de elaboração do produto, através de práticas inadequadas de higiene. Dessa forma, o objetivo deste estudo foi avaliar a qualidade e a segurança microbiológica na produção de queijos de média umidade. Amostras da matéria prima, dos manipuladores e do produto final foram coletadas em três laticínios situados na região sul do Rio Grande do Sul, com diferentes níveis de inspeção sanitária. A eficiência da pasteurização do leite também foi avaliada. Coliformes termotolerantes, estafilococos coagulase-positivos (ECP), Salmonella spp. e Listeria monocytogenes foram avaliados. As amostras de leite cru foram as que apresentaram os maiores níveis de contaminação, com enumeração de 1,1x105 número mais provável (NMP) mL-1 para coliformes termotolerantes, 4x105 unidades formadoras de colônia (UFC) mL-1 para ECP e a presença de Salmonella spp.. Contudo, apenas uma amostra de produto final estava em desacordo com o padrão regulamentar vigente, excedendo o limite permitido para ECP. A pasteurização do leite utilizado no preparo dos queijos foi eficiente em todos os laticínios, independentemente do nível de inspeção sanitária dos estabelecimentos. No entanto, houve contaminação pré e pós-pasteurização, demonstrando a necessidade de melhores práticas higiênicas, tanto na obtenção da matéria-prima, quanto na manipulação durante as diversas etapas de fabricação dos queijos.

ABSTRACT

Coagulase-positive staphylococci (CoPS) are opportunistic pathogens carrying various mechanisms of resistance that have a large number of virulence factors, and whose ability to induce illness is associated with the host. This study aimed to investigate the presence of environmental coagulase-positive staphylococci, their susceptibility profile, clonal relationship and ability to form biofilm. The 16S rRNA genes from CoPS isolates were analyzed, and their antibiotic susceptibility was evaluated using the agar dilution method in accordance with Clinical and Laboratory Standards Institute guidelines. The clonal profile was obtained by pulsed-field gel electrophoresis (PFGE) and biofilm formation was measured by a crystal violet retention assay. A total of 72 Staphylococcus spp. strains were isolated from air, metal surfaces, and nostrils from humans, dogs, cats, and birds. Three species were identified: Staphylococcus aureus (17%), Staphylococcus intermedius (63%), and Staphylococcus pseudintermedius (21%). Ninety three percent (93%) of the strains were resistant to at least one of 13 tested antibiotics. S. pseudintermedius strains were the only resistant ones to methicillin while most of these isolates were multidrug-resistant, had significantly higher ability to form biofilm and PFGE grouped into seven different patterns, without showing clonal dispersion among animals and environmental isolates. This study suggests that dogs, cat, and air are environmental sources potentially carrying multidrug-resistant S. pseudintermedius, which survives in different environments through biofilm formation and multidrug resistance, characteristics that can be transmitted horizontally to other bacteria and exacerbate the problem of antibiotic resistance in humans.


Los estafilococos coagulasa-positiva (CoPS) son patógenos oportunistas, portan varios mecanismos de resistencia, tienen un gran número de factores de virulencia y su capacidad para inducir la enfermedad está asociada con el hospedero. El objetivo de este estudio fue investigar la presencia de CoPS en el medio ambiente, su perfil de sensibilidad a los antibióticos, su relación clonal y su capacidad para formar biopelícula. De los aislamientos de CoPS se analizaron los genes 16S ARNr y se evaluó la sensibilidad a los antibióticos mediante el método de dilución en agar según el CLSI. El perfil clonal se obtuvo por electroforesis en gel de campo pulsado (PFGE) y la formación de biopelícula se analizó por retención de cristal violeta. Se aislaron 72 cepas de Staphylococcus spp. a partir de aire, superficies metálicas y narinas de humanos, perros, gatos y aves. Se identificaron tres especies: Staphylococcus aureus (17%), Staphylococcus intermedius (62%) y Staphylococcus pseudintermedius (21%). El 93% de las cepas fueron resistentes al menos a uno de 13 antibióticos probados. Los aislamientos de S. pseudintermedius fueron los únicos resistentes a meticilina y la mayoría fueron resistentes a múltiples fármcos, tuvieron una capacidad significativamente mayor para producir biopelícula y la PFGE los agrupó en 7 diferentes patrones, sin mostrar dispersión clonal entre los aislamientos de animales y de medio ambiente. Este estudio sugiere que los perros, los gatos y el aire son fuentes ambientales potencialmente portadoras de S. pseudintermedius resistente a múltiples antibióticos. Este agente sobrevive en diferentes entornos en virtud de la formación de biopelículas y la resistencia a múltiples antibióticos, características que pueden transmitirse horizontalmente a otras bacterias y, por ende, exacerbar el problema de la resistencia a los antibióticos en humanos.

ABSTRACT

No Brasil, o leite cru é um dos principais produtos agropecuários, devido principalmente ao seu elevado comércio e consumo. O crescimento desse comércio e consumo está relacionado com a importância econômica e nutricional que esse produto representa, pois, além de gerar empregos e renda, o leite desempenha um importante papel na alimentação humana. Uma das consequências do consumo do leite cru é a possibilidade de veicular doenças transmitidas por alimentos. Neste contexto, esta pesquisa teve como objetivo avaliar a contaminação do leite cru comercializado nas vias públicas do município de Açailândia/ MA Staphylococcus coagulase positiva. Foram analisadas cento e vinte amostras coletadas no período de março a outubro de 2014. Os resultados evidenciaram que 100% das amostras analisadas apresentaram contaminação por Staphylococcus coagulase positiva. A ocorrência de Staphylococcus coagulase positiva potencialmente enterotoxigênicos no leite cru representa um problema de Saúde Pública devido ao risco de causar intoxicação alimentar, além de ressaltar a necessidade de reavaliação dos padrões microbiológicos estabelecidos pela legislação brasileira para este produto.


In Brazil, the fresh milk is one of the main agricultural products, mainly due to its high trade and consumption. The growth of this trade and consumption is related to the economic and nutritional importance that this product is because, in addition to generating jobs and income, milk plays an important role in human nutrition. One of the consequences of the consumption of fresh milk is the possibility of vehicular foodborne illness. In this context, this study aimed to identify the presence of coagulase-positive Staphylococci in raw milk marketed informally in the municipality of Açailândia/MA. Were analyzed hundred and twenty samples collected in the period from March to October 2014. The results showed that 100% of the samples were contaminated by Staphylococcus coagulase positive. The occurrence of potentially enterotoxigenic Staphylococcus coagulase positive in fresh milk poses a public health problem because of the risk of causing food poisoning, and underline the need for reevaluation of the microbiological standards established by the Brazilian legislation for this product.

[...] As bactérias foram isoladas de amostras de leite compostas de todos os quartos mamários de cada vaca após descartar os primeiros três ou quatro jatos de leite. Para acessar os potenciais fatores de risco, características dos animais foram obtidas através de uma entrevista com os produtores. Os exames laboratoriais foram realizados de acordo com as recomendações do National Mastitis Council. Um total de 242 isolados foi obtido de 195 vacas a partir da amostra do rebanho total (251 vacas). A prevalência de infecções foi descrita em grupos de acordo com o perfil epidemiológico: bactérias ambientais, contagiosas e outras. Estas perfizeram 57,3 por cento, 26,3 por cento e 11,2 por cento, respectivamente, dos animais amostrados. Testes de suscetibilidade antimicrobiana contra 12 diferentes antimicrobianos foram realizados em 159 isolados. No total, 30 por cento dos isolados testados mostraram resistência a pelo menos três grupos diferentes de antimicrobianos e foram classificados como multirresistentes. Foram observadas as freqüências mais elevadas de resistência contra a ampicilina para os estafilococos coagulase-negativo, seguida de eritromicina para estafilococos coagulase-positivo e tetraciclina para estreptococos. A análise de regressão logística mostrou uma relação significativa entre a idade das vacas e a presença de estafilococos coagulase-positivo multirresistentes e distribuição de classes diferentes de bactérias nos diferentes estratos etários, o que sugere uma concorrência dinâmica ao longo do tempo (p < 0,05). Animais com três a quatro anos tiveram 13,7 vezes mais chances (IC95 por cento 1,4 - 130,2, p = 0,02) de ter estafilococos coagulase-positivo multirresistentes em comparação com aqueles com dois ou três anos. O tempo de exposição a agentes infecciosos e consequentes terapias sugere uma maior chance de colonização do úbere por patógenos resistentes devido à pressão de seleção repetida durante a vida.


[...] Bacteria were isolated from composite milk samples obtained from all quarters of each cow after discarding the initial three or four streams of milk. To access potential risk factors, animal characteristics were obtained through an interview with the producers. Laboratory tests were done according to National Mastitis Council recommendations. A total of 242 isolates was obtained from 195 cows out of 251 cows sampled. The prevalence of animal infections was described in groups according to the epidemiological profile: environmental, contagious and other bacteria. These were 57.3 percent, 26.3 percent and 11.2 percent, respectively of the sampled animals. Antimicrobial susceptibility tests against 12 different antimicrobials were performed in 159 isolates. Altogether, 30 percent of the isolates tested showed resistance to at least three different antimicrobial groups and were classified as multidrug-resistant. Higher frequencies of resistance were observed against ampicillin to coagulase-negative staphylococci, followed by erythromycin to coagulase-positive staphylococci and tetracycline to streptococci. The logistic regression analysis showed a significant relationship between age of the cows and presence of multidrug-resistant coagulase-positive staphylococci and distribution of different class of bacteria, suggesting a competition dynamic throughout the ages (p < 0.05). Animals with three to four years old had 13.7 times more chances (IC95 percent 1.4 - 130.2; p = 0,02) to have multidrug-resistant coagulase-positive staphylococci compared to those with two to three years. Time of exposure to infectious agents and consequent therapies suggests a greater chance of udder's colonization by resistant pathogens due to repeatedly selection pressure during lifetime.

The ripening process of Serro Minas cheese, one of the most popular cheeses produced with raw milk in Brazil, was studied over the course of 60 days of ripening during dry and rainy seasons. Brazilian legislation prohibits the production of cheese from raw milk unless it was submitted to a maturation period greater than 60 days. However Minas Serro cheese is sold within a few days of ripening. A total of 100 samples of Serro cheese were obtained from five farms; 50 samples were collected during the dry season (winter in Brazil) and 50 samples were collected during the rainy season (summer in Brazil). From each farm, ten cheeses were collected during each season after two days of ripening. Our results showed high levels of total and fecal coliforms at the beginning of the ripening period (approximately 4 Log MPN/g with 3 days of ripening) that decreased with 60 days of ripening reaching almost 1.5 Log MPN/g. Contamination by coagulase-positive staphylococci was reduced by the end of the ripening period. Salmonella spp. was not detected. The staphylococcal enterotoxins B and C were detected in 1% and 4% of the cheeses, respectively, after 30 days of ripening. These results suggest that the ripening process was not effective in eliminating staphylococcal enterotoxins from the cheese. However, none of the investigated strains of Staphylococcus spp. isolated from Serro cheese produced enterotoxins A, B, C or D. The high pathogen and coliform levels at the beginning of the ripening process for the cheese produced during both seasons indicate the need for improvement of the sanitation of the manufacturing conditions.

ABSTRACT

O objetivo deste trabalho foi identificar espécies de Staphylococcus (n=100) isoladas de mastite em rebanhos bovinos do Estado de Minas Gerais. Para esta finalidade foram utilizadas reações de PCR empregando oligonucleotídeos iniciadores descritos anteriormente para amplificar genes específicos de S. aureus (femA), S. intermedius (rDNA 16S) e S. hyicus (rDNA 16S-23S) e o sequenciamento do rDNA 16S. De acordo com as reações de PCR, 83 isolados foram identificados como S. aureus, 13 isolados como S. intermedius, dois como S. hyicus e dois isolados não foram identificados. Foram submetidos ao sequenciamento do rDNA 16S seis isolados identificados como S. aureus e os 17 restantes. Os seis isolados identificados como S. aureus confirmaram essa identificação. Dos outros 17 isolados, 13 foram identificados como S. chromogenes e quatro como S. hyicus, com similaridade igual ou superior a 99%. Baseando-se nos resultados da reação de PCR do gene femA e do sequenciamento do rDNA 16S, foram identificados 83 S. aureus, 13 S. chromogenes e quatro S. hyicus. Neste estudo os oligonucleotídeos iniciadores empregados na reação de PCR para S. intermedius não foram específicos, pois amplificaram também S. chromogenes; e os empregados na reação de PCR para S. hyicus não foram sensíveis, pois falharam na identificação de dois isolados de S. hyicus. A identificação definitiva das duas últimas espécies somente foi possível pelo sequenciamento do rDNA 16S.


The objective of this study was to identify the species of 100 isolates of Staphylococcus from mastitis in dairy cows from herds located in the state of Minas Gerais, Brazil. PCR reactions were carried out using specific primers described previously for S. aureus (femA gene), S. intermedius (16S rDNA) and S. hyicus (16S-23S rDNA spacer region). In addition, products of amplification of variable regions of the 16S rDNA gene of the strains were sequenced. According to the results of the PCR, 83 strains were identified as S. aureus, 13 as S. intermedius, two as S. hyicus and two isolates were not identified. The sequencing of 16S rDNA was applied to 23 strains identified by PCR amplifications: six S. aureus and the strains identified as S. intermedius (n=13), S. hyicus (n=2) or not identified (n=2). The sequencing of 16S rDNA confirmed the six strains as S. aureus. The others 17 strains were identified as S. chromogenes (13 isolates) and S. hyicus (four isolates). Each sample was related to a specie according to the smallest E-value and highest similarity (≥ 99%). The identification of S. hyicus and S. chromogenes was accomplished only by 16S rDNA sequencing.

Os serviços de alimentação coletiva têm se expandido em todo o mundo, sendo os restaurantes do tipo self-service a preferência atual dos consumidores. Considerando-se a importância da qualidade higiênica dos alimentos para a saúde da população, foi investigada a microbiota presente em alimentos prontos para o consumo. No segundo semestre de 2008, foram coletadas 20 amostras de refeições, à base de carne, prontas para o consumo, em diferentes restaurantes self-service da cidade de Araçatuba/SP. As análises bacteriológicas realizadas seguiram as metodologias convencionais e os resultados foram comparados com os padrões regulamentados pela legislação brasileira vigente. Das amostras analisadas, 90% foram positivas para coliformes a 35ºC. Coliformes a 45°C foram detectados em 55% das amostras e, destes, em 63,63% foi confirmada a ocorrência de Escherichia coli. A presença de estafilococos coagulase-positiva foi verificada em 10% das amostras; Salmonella spp. e Bacillus cereus não foram detectados. Não foram pesquisados os clostrídios sulfito redutores a 42ºC. Este estudo aponta a necessidade de atenção rigorosa quanto às condições sanitárias de preparo e exposição dos alimentos prontos para consumo, uma vez que a ingestão de produtos contaminados constitui um potencial risco para a saúde pública.


Collective food services have been increasing worldwide, and the self-service restaurant has been the current preference by consumers. Considering the importance of hygienic quality of food, the microbiologicalcomposition of ready-to-eat food was assessed. In the second semester of 2008, 20 samples of meals, mainlymeat-based foods, were collected from different self-service restaurants in Araçatuba city, SP. Bacteriologicalanalyses were performed following the conventional methodologies, and the results were compared with thestandards established by the effective Brazilian legislation. Coliforms at 35ºC were detected in 90% of analyzedsamples. Coliforms at 45°C were found in 55% of the samples and, among these, in 63.63%, the occurrence of Escherichia coli was confirmed. Coagulase-positive staphylococci were detected in 10% of samples and no sample showed Salmonella spp. or Bacillus cereus contamination. Sulfite reducing clostridia at 42oC were not investigated in this study. These findings indicate the need for a rigorous approach for improving the sanitary conditions during preparation and presentation of ready-to-eat food, as the consumption of contaminated products represents a potential risk to public health.

ABSTRACT

Sixty-five strains of coagulase positive staphylococci (Staphylococcus aureus, S. intermedius and S. hyicus) were identified at species level by PCR amplification of the coa gene, specific for S. aureus, and of the nuc gene, specific for S. intermedius and for S. hyicus.


Sessenta e cinco cepas de estafilococos coagulase positiva foram identificadas em nível de espécie, através da amplificação, por PCR, de seqüências do gene coa, específicas para S. aureus, e do gene nuc, específicas para S. intermedius e para S. hyicus.
